The table that follows is a guide to the activity of some of our restriction enzymes in One-Phor-All Buffer PLUS (OPA+). Enzymes not listed in the table have not been tested for activity in OPA+ buffer. For each enzyme in the table the following pieces of information are provided:

The optimum concentration of OPA+ buffer - the concentration at which the enzyme's activity is highest (the buffer is a 10× concentrate1).
The enzyme's activity in the optimum concentration of OPA
+ buffer as a percentage of its activity in the assay buffer listed on the Certificate of Analysis.

Percentages are designated as follows: ++ = 50-99%; +++ = 100%; ++++ = 101-199%; +++++ > 200%

1   OPA+ buffer (27-0901-02) will be provided free of charge upon request. Please inquire.
2   With Bgl I, addition of KCl to a final concentration of 100 mM KCl is required.
3   With EcoR I and Nco I, star activity may be observed in 1× OPA+ when DNA is digested for longer than 1 hour. Star activity can be eliminated during extended incubations by using 2× concentration.
4   With Not I, addition of Triton X-100 to a final concentration of 0.01% (w/v) is required.
5   With Pvu II, addition of NaCl to a final concentration of 50 mM is required.
